    Background

    E7820 is an anti-cancer sulfonamide that acts as an angiogenesis inhibitor, known to degrade the RNA splicing factor RBM39 (1). It is understood that patients with acute myeloid leukemia (AML) or myelodysplastic syndrome (MDS) can exhibit mutations in RNA splicing factor genes, including RBM39, which is crucial for AML cell survival (2). By acting as a splicing factor inhibitor, E7820 encourages RBM39 proteasomal degradation, which can help suppress splicing events that may contribute to unchecked cancer cell growth (2). It is also thought that the effect E7820 has on splicing factors could potentially influence integrin expression and/or function, specifically, integrin α2 (1). Because integrins are involved in cell migration, splicing factor degradation can play a role in altering the manner in which cancer cells migrate (3). The E7820 IC50 value varies depending on the context in which the drug is studied but can range from 1-5 µM for splicing factor inhibition (2).
